5-Amino-1MQ is one of those compounds that often gets grouped in with peptides, but chemically, it isnβt a peptide at all.
Its full name is 5-amino-1-methylquinolinium, and it belongs to a class of small molecules developed to inhibit an enzyme called nicotinamide N-methyltransferase, or NNMT.
And NNMT is where the story gets interesting.
NNMT interacts with some extremely important metabolic chemistry.
It uses nicotinamide β a form of vitamin B3 β and a molecule called SAM, or S-adenosylmethionine, in a methylation reaction.
Why does that matter?
Because nicotinamide is also part of the pathway the body uses to make NAD+ β one of the most important molecules involved in cellular energy metabolism.
That led researchers to ask a fascinating question:
What happens if you inhibit NNMT and change the way those metabolic resources are being used?
Interest in that question exploded after a landmark 2014 study in Nature found that reducing NNMT activity in the liver and fat tissue of mice altered cellular metabolism and protected the animals against diet-induced obesity.
Researchers subsequently developed small molecules capable of directly inhibiting NNMT.
One of those molecules was 5-Amino-1MQ.
And this is where the compound started attracting serious attention.
