DIHEXA

Dihexa is one of the more unusual compounds in peptide research because its story begins with Angiotensin IV, a naturally occurring peptide researchers had already been studying for its relationship with learning and memory.

The problem was that Angiotensin-IV-based compounds were difficult to work with: they could be broken down quickly and had poor ability to get across biological barriers.

Researchers at Washington State University began modifying the active portion of Angiotensin IV, eventually developing Dihexa as a much more metabolically stable research compound designed to reach the central nervous system more effectively.

And that is where things get interesting.

Rather than simply looking at whether a compound temporarily changes neurotransmitter activity, much of the early Dihexa research focused on something more structural:

The connections between neurons themselves.

Researchers investigated Dihexa in relation to:

• Dendritic spine formation
• Synaptogenesis — the formation of new connections between neurons
• Learning and memory
• Spatial memory
• Neuroplasticity
• Neuronal survival and signaling
• Cognitive impairment models
• Alzheimer’s-related animal models
• Cellular pathways involved in brain repair and adaptation

In early experiments, Dihexa was associated with increased hippocampal synaptic connectivity and improvements in memory tasks in  whose cognition had been experimentally impaired.

That distinction matters.

Researchers weren’t simply asking, “Can this make someone more alert?”

They were investigating whether damaged or poorly connected neural networks could actually become better connected.

More recent research in an APP/PS1 mouse model of Alzheimer’s disease also reported improvements in spatial learning and memory along with changes in neuronal and synaptic markers. Researchers additionally investigated the PI3K/AKT signaling pathway, inflammation-related signaling, and neuronal survival.

Dihexa has also been studied in connection with the HGF/c-Met system — hepatocyte growth factor and its Met receptor, a pathway involved in cell growth, survival, migration, and nervous-system development.

That proposed mechanism is still an evolving area of research rather than settled science, but it helped make Dihexa an especially unusual compound because researchers were looking at structural neuroplasticity and synaptic connectivity, not simply short-term stimulation.

That’s really the fascinating part of Dihexa:

It represents a completely different research question.

What if cognitive decline isn’t approached only by changing brain chemistry, but by studying ways the brain may form and strengthen connections between neurons?
